Ventricular mass was increased in XRT-only animals compared with controls (87 mg versus 53 mg, p=0.0001), suggestive of pathological left ventricular hypertrophy (Figure 2C). This was not observed in XRT+Entresto animals (59 mg) however. Similarly, end-diastolic volume, a marker of systolic pump function, was significantly increased in irradiated animals compared with controls (68 μL versus 37 μL, p=0.0001) and this change was also not seen in the XRT-Entresto group (Figure 2D).

Differences between control and Entresto-only arm were not statistically significant for any parameter at either timepoint, and no adverse effects were noted in the Entresto-only arm at the dose utilised.

Conclusion:

Entresto was well tolerated and attenuated the RC phenotype in these early systolic function and structural readouts. Changes detected by global longitudinal strain were more pronounced than classic systolic parameters at 10–20 weeks, indicating that this parameter may have utility as a reliable early biomarker of radiation cardiotoxicity. Further studies to investigate Entresto as a novel radioprotectant are warranted from the preliminary analysis presented.
