However, for both GnRHR agonists Leu and Trp, such positive effect emerged to be mainly associated to the lower doses only, while the higher being only minimally or not at all able to be beneficial, as apparently indicated by the heterogeneity of the results. These effects were also confirmed by the analysis of bladder structure in terms of fibrotic tissue limitation and urothelium integrity conservation. On their whole, our data indicate that the androgen depletion therapy can be beneficial even at lower doses, which are likely to cause less side effects.

Conclusion:

Our pre-clinical model of RT-induced bladder damage allowed to test and compare the effect of three different drugs in the context of the androgen-deprivation therapy. They all resulted effective to counteract the radiation induced functional decline of the bladder, with main differences due to the selected doses, suggesting a potential role as therapeutic solutions to be further developed in clinical practice.
