Conclusion:
Conclusion: We show that the inhibition of ATM or ATR results in increased radiosensitivity and leads to downstream DNA repair deficiency as reflected in the decrease in yH2AX and 53BP1 presence. The increased STING phosphorylation after ATM inhibition plus irradiation of DU145 cells implies that the combination of irradiation and ATM inhibition could result in increased immunogenicity. This highlights the complex interactions between the DDR machinery and immune related mechanisms like STING activation. We will continue to investigate these interactions more in-depth, with focus on the cGAS-STING pathway and the possible increase in immunogenicity of prostate cancer cells after ATM/ATR inhibition.
