Similarly, SK-LMS-1 sarcoma MDTs irradiated with 8Gy showed a reduction in viable cells, nuclear area (data not shown), and Ki-67 expression and an elevation in the level of g-H2AX foci (Figure 2.A and C). Figure 2.B illustrates some representative images in each condition. Furthermore, ctDNA as a proxy of cell death was assessed (Figure 2.D). RT alone did not significantly increased ctDNA release but adding AZD4320 caused a spike in it 1 and 2 days after AZD4320 treatment. AZD4320 alone and RT+ AZD4320 showed the same level of ctDNA release. However, as discussed, irradiated MDTs contained less viable cells, suggesting that there were proportionally more cell deaths and ctDNA release in the combination therapy.
