AIMS AND OBJECTIVES

1. To assess and compare the radiological responses.

2. Assessment of toxicities (acute and late).

Material/Methods:

Patients with newly diagnosed, biopsy proven high grade glioma were randomly assigned in two arms-

Experimental arm (ARM A)

Patients in arm A were treated by post-operative TMZ 150mg/m 2 per oral Day 1 to Day 5 at 28 days interval for 2 cycles, then concomitant chemoradiation with TMZ 75mg/m 2 per oral daily for 42 days and external beam radiation therapy using conventional fractionation up to total dose of 60Gy followed by 4 cycles of TMZ150mg/m 2 per oral Day 1 to Day 5 at 28 days interval.

Control arm (ARM B)

Patients in arm B were treated by post-operative concomitant chemoradiation with TMZ 75mg/m 2 per oral daily for 42 days and external beam radiation therapy using conventional fractionation up to total dose of 60Gy followed by 6 cycles of TMZ 150mg/m 2 per oral Day 1 to Day 5 at 28 days interval.

Response assessment and toxicity study were done for both the arms

Results:

A total of 60 patients underwent randomization, 30 patients in each arm. 53.3% patient in arm A and 50% patient in arm B were in 51-60 years age group.

In arm-A, 5 (16.7%) patients had COMPLETE RESPONSE (CR) after 6 Weeks, 4 (13.3%) patients had CR after 3 Months, 3 (10.0%) patients had CR after 6 Months. In arm B, 2 (6.7%) patients had CR after 6 Weeks, 1 (3.3%) patient had CR after 3 Months. In arm A 23 (76.7%) patients had PARTIAL RESPONSE (PR) after 6 Weeks, 24 (80.0%) patients had PR after 3 Months and 24 (80.0%) patients had PR after 6 Months. In arm B 26 (86.7%) patients had PR after 6 Weeks, 25 (83.3%) patients had PR after 3 Months, 26 (86.7%) patients had PR after 6 Months.

In arm A, 10 (33.3%) patients had ANAEMIA Grade 1 and 11 (36.7%) patients had ANAEMIA Grade 2 at WEEK1. In arm B 4 (13.3%) patients had ANAEMIA Grade 1 and 18 (60.0%) patients had ANAEMIA Grade 2 at WEEK1.

In arm-A, 11 (36.7%) patients had DIARRHOEA Grade 1 and 7 (23.3%) patients had DIARRHOEA Grade 2 at WEEK 1. In arm-B, 14 (46.7%) patients had DIARRHOEA Grade 1 at WEEK 1 and 6 (20.0%) patients had DIARRHOEA Grade 2 at WEEK 1.

In arm-A, the mean SURVIVAL DURATION (in days) (mean ± SD) of patients was 280.0 ± 99.8.
