environment for tumor progression. To identify cellular mechanisms, we established a 3D co-culture model with HNC cells and monocytes THP1. The 3D culture unlike the monolayer culture of cells can mimic different features of human tissues, including cellular organization, cell-cell and cell-extracellular matrix (ECM) interactions essential to understand the mechanisms underlying disease progression. Materials and Methods We choose the Liquid Overlay Technique (LOT) which relies on the self-aggregation of cells in low adhesion plates with round-bottomed that promotes cell-cell interactions and therefore enhance the formation of spheroids. First, we have characterized and optimized the seeding densities of cancer cells (FaDu HPV- HNSC and 93VU HPV+ HNSC) spheroids in 96-well plates at different days. Next, we enhanced the spheroid system with the addition of THP1 to recreate the environment, by a hybrid spheroid system, more accurately with different ratios of cancer cells and macrophages ranging from 1: 1 to 1:5. Spheroids were collected and embedded in paraffin to investigate the profile of monocyte/macrophage by immunohistochemistry. Analysis of the expression of defined cell surface markers and soluble factors was characterized by Real-time qPCR and flow cytometry in comparison with the 2D co-culture realized by exposing THP1 cells to conditioned medium prepared from cultures of lines 93VU and FaDu. Results The first results allowed us to select the number of cells to seed for each cell line as well as the day in which we must collect our spheroids before they lose their shape. We notice that the spheroids can be formed from day 2 until the augmentation of the necrotic core in the middle at day 6. Using this model, we will analyze the influence of tumor cells on monocytes and their immunosuppressive phenotype. We will show that the THP-1 infiltrate the tumor tissue, induce the production of immunosuppressive cytokines and polarize into an M2-like macrophage phenotype expressing CD68, CD163 and CD206 corresponding to the TAM phenotype.

Conclusion A co-culture based on spheroids can be an advantageous tool for understanding the cross-talk between immune and cancer cells and could be applied in the evaluation of drug and irradiation treatments on HNC cells.

PO-0155 Health related QLQ in Head & cancer survivors - a post treatment review in A single centre

Purpose or Objective - Among Cancer Patient the QLQ of HN cancer survivors is an important part. With improvement in the treatment technique the survival rate has improved. Post- treatment they have enfeebling problem with swallowing, speech, & hearing, and psychological effects of loss function and body image. In this study we have consider the worst-case scenario to analyze there QLQ. Materials and Methods 622 Patients were screened, coming for follow-up since 2011. Among the survivors there 15% were Female and 85% Male, with most common site of malignancy among them was oral cavity (71%). Patient were treated with Chemo therapy, Surgery & Radiotherapy. These patients were analyzed on QLQ-35, QLQ-30 and HADs (Hospital Anxiety & Depression Scale). Results On HAD scale Patients feeling anxious (48%) were higher than feeling depressed (38%). Patients reported sore in mouth (41%), had issue in swallowing solid diet (58%), had decreased in taste (43%), had issue in speech while talking to others (47%), lacked social contacted due to their appearance (49%) and had sexually enjoyed less (25%)on HN-35 scale. While on HN-30 patients who had notified disturbance in social functional as their disease had inferred the family life were 51%, in cognitive functional patients reported high stress were 49%, followed by emotional functional disturbance as they had difficulty in concertation were 49%. On symptom Scale patients who felt weak were 42%. Conclusion Quality of Life is impaired in all patients treated with chemo-radiotherapy. IN our centre we tried to asses this by using EORTC SCALE and HAD scale. It was different for different sites and depends upon the treatment received and span of survival.
